dc.description.abstract | This study aimed to investigate some biological activities and antiinflammatory potential of polysaccharide isolated from Leucoagaricus leucothites in LPS-activated BV-2 cells. Here, we investigated the antioxidant and antimicrobial potential of polysaccharide extract of L. leucothites (LLP) and the effect of LLP on lipopolysaccharide (LPS)-induced inflammatory response and the underlying molecular mechanisms in BV-2 cells. Chemical analysis of LLP were determined by high-performance liquid chromatography (HPLC). Cytotoxicity and productions of TNF-alpha and IL-6 were determined by ELISA. In addition, intracellular mitogen-activated protein kinase (MAPK) signaling pathway was analyzed by Western blot and cyclooxygenase-2 (COX-2) and nitric oxide synthase (iNOS) expression were measured by real time-PCR. LLP inhibited LPS-stimulated IL-6 and TNF-alpha. In addition, the inhibitory action of LLP not only suppressed COX-2 and iNOS, but also inhibited the activation of p38/ERK MAPKs. Our data suggested that LLP exhibit anti-inflammatory activity that might contribute to the prevention of chronic inflammatory diseases. | en_US |
